Neal Oliver (James Marsden) is een jonge tekenaar, wiens vader liever ziet dat hij in Oxford gaat studeren. Hij wordt in een bizar en onverwacht avontuur gestort wanneer hij de sadistische half-gnoom O.W. Grant (Gary Oldman) ontmoet. Deze biedt Neal aan een wens te vervullen en de dromer Neal gaat keihard voor de bijl. Hij wenst antwoorden op al zijn vragen! O.W. Grant stuurt hem op pad over de niet bestaande Interstate 60 naar het niet bestaande Danver. Onderweg hoopt hij dat de billboards hem naar de vrouw van zijn dromen leiden maar of zijn wens hier echt in vervulling gaat...
